Abstract

Colorectal cancer (CRC) remains a major global health challenge, and the emergence of treatment resistance poses a significant hurdle in achieving optimal therapeutic outcomes. Treatment resistance in CRC can manifest through various mechanisms, including genetic mutations, microsatellite instability, epithelial-to-mesenchymal transition, cancer stem cells, apoptosis resistance, and interactions within the tumor microenvironment. Addressing these complexities necessitates innovative strategies to enhance treatment efficacy. Targeted therapies, immunotherapies, combination therapies, and personalized medicine strategies are good examples of promising avenues and frontiers in tackling CRC treatment resistance. Advancements in understanding the tumor microenvironment guide the development of therapies that modulate interactions between cancer cells and their surroundings. These noninvasive methods provide valuable insights into evolving tumor landscapes, facilitating adaptive treatment strategies. As the field of CRC treatment evolves, integrating these emerging approaches can transform the landscape of therapeutic interventions. By understanding the underlying mechanisms of resistance and tailoring treatments based on individual tumor characteristics, clinicians are poised to enhance treatment outcomes and improve the prognosis for CRC patients. Continued research and clinical trials will be essential to validate the efficacy and safety of these emerging treatment approaches in diverse patient populations. This chapter highlights the latest breakthroughs and emerging treatment approaches for CRC resistance. We delve into novel drug targets, experimental therapies, and cutting-edge research that offer hope for patients who have exhausted traditional treatment options.
