Effect of hydroxylated and methoxylated coumarins on the regulatory properties of phenylalanine ammonia-lyase from Citrus sinensis

Abstract

The effects of hydroxylated and methoxylated coumarin derivatives, occurring in γ-irradiated citrus flavedo, on phenylalanine ammonia-lyase from Citrus sinensis were investigated. Umbelliferone and scopoletin inhibited the enzyme by increasing the negative cooperativity between the substrate binding sites. Esculetin, scoparone and fraxetin activated the enzyme, with fraxetin effecting a decrease in negative cooperativity. The coumarin derivatives affected the Vmax but not the Km kinetic parameter.
